Core B: Biochemistry and Structural Modeling Core
The Biochemistry and Structural Modeling Core focuses on determining the biochemical composition and atomically detailed structural models of apoE/lipoproteins containing the apoE2, apoE3, and apoE4 isoforms. One hurdle is that the field currently lacks a detailed understanding of the biochemical and structural properties of different apoE isoforms in their native, lipid-bound states referred to as apoE/lipoproteins. It is also unclear whether these properties vary with cell-type, or if/how apoE/lipoproteins interact with amyloid-β (Aβ). To shed new light on these questions, Core B supports other U19 Projects/Cores by 1) isolating and characterizing apoE/lipoproteins, 2) providing structural modeling services, and 3) analyzing the data generated to identify biochemical and structural signatures that predict the impact of different apoE isoforms on aging and AD.
